CAGRILINTIDE + SEMAGLUTIDE

CAGRILINTIDE + SEMAGLUTIDE

What it is

The body's two satiety pathways, in a single vial. It combines the amylin analog with the most studied GLP-1 analog in the world. In the literature it is known as CagriSema.

The body has two different systems for saying “enough”: incretins, which come from the gut, and amylin, which comes from the pancreas. Each one works on its own. This combination activates both at once, which is why the combined effect is greater than that of either one separately.

What it is used for

  • • Research on weight control through two simultaneous pathways
  • • Satiety, appetite and gastric emptying
  • • Studies of the combination versus each component separately
  • • Body composition and cardiometabolic risk factors

How it is used

Cycle
Continuous until target
Break
Not required
Route
Weekly subcutaneous

Lyophilized powder with both peptides already combined. It is reconstituted with bacteriostatic water, swirled without shaking, and stored between 2 and 8 °C. In clinical trials it is administered once a week with gradual escalation [1].

What has been observed

Results described in the published scientific literature. They are not directions for use or health recommendations.

  • • In the phase 3 REDEFINE 1 trial (3,417 adults, 68 weeks), the mean weight reduction was 20.4% versus 3.0% with placebo [1]
  • • Participants reached weight loss targets of 5%, 20%, 25% and even 30% of body weight far more often [1]
  • • For reference, semaglutide alone reached 14.9% at 68 weeks in STEP 1, and cagrilintide alone 10.8% in its phase 2 [4][5]
  • • Digestive effects were frequent (79.6% versus 39.9% with placebo), but mostly transient and mild to moderate in intensity [1]
  • • An analysis of REDEFINE 1 also described a reduction in blood pressure [7]
  • • There are also phase 3 data in people with type 2 diabetes [2]

How it works

Semaglutide — GLP-1 receptor agonist

An analog of the incretin GLP-1 with substitutions that extend its half-life enough to allow weekly administration. It acts on the hypothalamic appetite centers and on glucose-dependent insulin secretion [4].

Cagrilintide — amylin analog

A long-acting amylin analog designed to resist degradation [6]. It acts on amylin and calcitonin receptors in the area postrema and slows gastric emptying [5].

Why they add up

Incretins and amylin are independent signaling systems that converge on the same behavior: stopping eating. The phase 1b trial already described an additive effect, and phase 3 confirmed it versus each component separately [3][1].

State of the evidence

Best in its category. Phase 3 published in The New England Journal of Medicine with more than three thousand participants, additional data in type 2 diabetes and secondary analyses on blood pressure [1][2][7]. Even so, the combination is not approved as a medicine, and the material we offer is research grade, not pharmaceutical grade.
